Study question
This study aimed to predict the parcentage of clinical pregnancy by comparing time to cleavage using EmbryoScope based on Meseguer report.

Summary answer
It turned out the analysis of time to 2-3 cells, 3-4 cells, from insemination to five cells as shown by EmbryoScope are related to parcentage of clinical pregnancy except 2-3 cells.

What is known already
The outcome of embryo using Embryoscope is connected between parcentage of clinical pregnancy and time to cleavage after embryo transfer (Meseguer et al.,2011). We explored if the standard is appropriate by investing Japanese cases.

Study design,size,duration
 In this clinical retrospective study, we analyzed 48 couples underwent ICSI(59) led to an embryo transfer from December 2012 to Deptember 2014 at our clinic.

Participants/materials,setting,methods
EmbryoScope (Unisense Fertilitech,Denmark),images were acquired every 15 min for each embryo. Target couples is maternal age≦38 and OPU≦3. All transfers were frozen cycles and embryos were transferred at either cleavage or blastocyst stages. The specific time was determined as follows: from 2-3cells (cc2), from 3-4 cells (s2), from insemination to five cells (t5). We compared the percentage of the in-range group (w/i group) and the out of range group
(w/o group) that achieved clinical pregnancy by the chi-square test.
 

Main results and the role of chance
The cc2 w/i group (11.9h<) is 37.5% and w/o group(11.9h≧) is 47.4%. There was no difference in time in cc2 between the w/i and w/o groups. The s2 w/i group (0.76h<) is 56.5% and w/o group(0.76≧) is 30.6%. The w/i group had a significantly higher ratio of obtaining clinical pregnancy than the w/o group (p=0.048). The t5 w/i group (44.8–56.6 h after insemination) is 51.4 and w/o group(outrage) is 25.0%. The w/i group had a significantly higher ratio of obtaining clinical pregnancy than the w/o group (p=0.042).
 

Limitaions, reasons for caution
We can get this result by limiting age and number of OPU. After this, we want to get new markers including age and number of OPU by being increased cases.

Wider implications of the findings
The outcome of embryo using Time-lapse System is connected between parcentage of pregnancy after embryo transfer and time to cleavage (Meseguer et al.,2011),and our study said s2 and t5 is important markers. We can select high-quality embryo for transfer by compareing the time of cleavage or blastocyst.
